You could read The Art Of Computer Viruses Research and Defense, it is a good book to know the history of malwares.


Then, vxheaven.org is really a good reference, look at the ezines section where you can find sources, most of them are written in C or Assembly. 


I suppose that you are targetting Windows OS, in this case, you should also have a good background on the PE format and the Win32 API / Windows Internals.
